The aim of this experiment is to invesitigate the effect of Bile salts on Lipase activity.
Equiptment And Materials
* establish Tube Rack with 3 Test Tubes
* 3 5ml Syringes
* 4 100ml Beakers
* Dropping Bottle of Universal Indicator
* Stopclock
* optic Protection
* Marker Pen
* Water Bath set at a continuous Temperature of 37?C
* 200ml Full Cream Milk
* 100ml of 3% Bile Salt Solution
* 50ml of 0.1M Sodium Carbonate Solution
Method
1. Collect all materials indicated above.
2. using marker pen, label the examination tubes with A, B and C.
3. correspond 5ml(5000µm) of draw to each test tube.
4. To test tubes A and C cast up 1ml(1000µm) of 3% bile salts solution. To test tube B add 1ml(1000µm) of Distilled Water.
5. Add 1cm(1000µm) of oecumenical indicator to each test tube.
6. Add 0.1M sodium carbonate solution slowly drop by drop until the contents of each tube are at pH7 (pale green in colour).
7. Add 2ml(2000µm) of the 5% Lipase solution to test tubes A and B and 2ml(2000µm) of distilled water to test tube C.
8. gamy shake the contents of the tubes so that the contents are mixed.
moorage tubes into the water bath set at a constant temperature of 37?C.
9. Take a note of the colour of the universal indicator in each test tube at the start and after a period of 1 hr. geminate the colour with the pH colour chart to obtain pH values. Put results into a table and record class results also.
